At the moment (and it usually changes very frequently), my favourite is Streets of Philidelphia by Bruce Springsteen. It is kind of depressing, but I love it.
When you feel down, it sort of makes you feel that you're not alone, and everything's going to be okay.
Oh, and I love This Womans Work, by Kate Bush, she has a fabulous voice and the song is quite inspirational, for want of a better word.
Very tough question to answer with just one song, but if I had to pick only one, then I would pick "Nessun Dorma" by Pavarotti. 2:33 of absolute beauty that will pull all the strings of your heart, and you don't have to know Italian to understand the song's meaning.

Hey ! Ice man, Good answer! I recently seen a t.v. special with these two...And it covered how BB & a bunch of old timers were his inspirations...Going back as far as Muddy Waters, Johnson...& his love for the original steel guitar...how it inspired his works through his whole life. It was great!
