my imidiate family (parents, sibs, nieces and nephews) no... never... aunt/uncle/cousin... i could, but i doubt i would.

If a family member was trying to kill me for some unjustified reason, and I feared that they had the means at hand to do it, I think I could. However, I've never been in that situation so I'm not sure. However, if you mean a situation like where my wife gets mad at me and tries to hit me then ultimately I'd just leave until she cooled off. After that I'd try to talk to her about it when she had enough time to cool off a little.

in self defense well thats what i would tell them in court... honestly one time i was gonna kill my mom and drink her blood so i could claim insanity and before you say anything shes a druggie who calls me a slut and a bitch all the time for no reason at all and though the beatings have stopped is hell to live with

Yes - when it comes to self-defense, any aggressor should be stopped. It may be unpleasent or traumatic, sure, but allowing yourself to be killed isn't ethical, it's just uncourageous.
