Let's see... an alien symbiont wants to meld with you and start taking over your life. You (Spider-Man) figure out to get it off you and it's kinda peeved at you and wants revenge. The symbiont then melds with someone who already doesn't like you (for stealing his job and the girl he likes), to double up on the hate. Hope this helps.

Eddie was the one who resented/hated parker first and the symbiote hated and knew who spider man was because the symbiote had become attached to Peter and was just as dependent and attracted to Peter's power as peter was to the symbiote and it's abilities...they had a symbiotic relationship! Mix the symbiotes hatred for being separated from peter with Eddie's hatred for Peter (especially after discovering he was also spider-man) and there you go!
