Though I haven't found a definitive answer, my guess is that it is derived from the fact that you would have to be the stupid one to wear the back half of the costume. :)

Mules are the offspring of a male (jack) ass and a female (mare) horse (from dictionary.com). So a horse's ass would be a male donkey kept to impregnate mares - like a gigolo.

It is actually a "horse's arse". In pantomime, the most ignominious part was playing the backside of a panto horse. So, if you were called a "horse's arse", you were as low as you could get.

not sure of origin but used in comedy, as in [ you sir, are a horses ass] very funny, python i think.

Not for sure, but they use a horse's body parts for a few things, such as, hung like a horse!
