They were punished by being thrown out of the garden and becoming mortal. As a result of leaving the garden, they had to start working for their own living. As a result of becoming mortal they became subject to injury, disease, sorrow, and eventually death.

By sending them out of the Garden is the simplest answer, but there is a sense where they punished themselves. They had the choice to obey or rebel. They chose rebellion (ie they wanted to be "gods") and this act set in place a chain of events. It put a schism between humans and the created order (which formerly had been in harmony- this led to all the diseases, deforestation, soil erosion, pollution, global warming etc) it put a schism between humans and other humans (leading to war, hatred, disputes, abuses etc) and worst of all a schism between holy God and rebellious humans (leading to physical and moral death). God, being holy, cannot abide sin in HIs presence, so He symbolised this by putting Adam and Eve outside the Garden and closing access to it. They had to live with the consequences of their actions. HOWEVER, from the beginning God put a rescue plan in place. He spoke of it first to Eve in Ch 3 of Genesis: "I will put enmity between him (the snake's- Satan's) and your seed. He will bite his (your seed's) heel but your seed will crush his head." This is the first reference to the coming of a Saviour who would crush sin and restore the harmony between humans and their God. We even see Eve's hope, at the birth of Cain, that this child might be the one. She, sadly, could not see that her actions and those of her husband had much bigger repercussions, but she still had hope. She trusted, once again, in the promises of God

Wasn't Adams punishment having to get everything from then on by the sweat of their brow and part of Eves punishment that she would give birth in pain, desire for her husband and be ruled over by him 3:17-19. Evedently there were two trees in the garden. The tree of the knowledge of good and evil and the tree of life. Noting the lack of prohibition they were free to eat from the tree of life but never did. God not only removed them as part of the punishment but also that neither they or anyone else having the knowledge of good and evil might eat from the tree of life and live forever.

He kicked them out of paradise, took away their chance at immortality, made Adam work for a living, gave Eve cramps and made her subject to Adam, and put enmity between them and the snake (Gen. 3:14-19). He also allowed their sin to be passed onto their ancestors, which made their relationship with God very difficult and tedious until Jesus came along.
