If I am Canadian and in the U.S. on an H1-B Visa, and I married an American, would I still be able to use that visa? If not, how long would it take me to be able to work in the U.S.?

Yes, you may as long as it is valid, but I suggest that upon marriage you and your wife file for an adjustment of status while the visa IS valid and get a conditional greencard.
