I love old movies, and the toughest woman was Katherine Hepburn. She took control at a time when women were portrayed in long skirts and frail. Not Kate, she played hard as nails characters who stood nose to nose or above with her male counterparts. She rarely wore makeup and was always in pants. Some of my favorite movies with Katherine: Holiday, Bringing Up Baby, Woman of the Year and The Philadelphia Story.

Elvira Mistress of the Dark, she's hella funny and is quick with her witty snaps and comebacks to tear anyone down.
She can make love spells and make a whole town bang each other in public. She also controls the teenage and male population with her big boobies
2nd Runner up would have to be Mila Jovavich, she just kicks major ass in all her recent films and also has some good phrases she says right before decimating her foe.

Probably Jennifer Connelly in "A Beautiful Mind." She plays the role of a strong, intelligent, and tough willed young lady that helps her husband through his hardships and still doesn't give up. Too bad I forgot her name in the movie. lol.
