The Greeks Aristotle and Plato wrote papers on the subject so I'd guess the search for the basic building block has been going on a lot longer than that.

Since the beginning of philosophy (at least written down) by early greeks. Lots of early greeks thought the basic building blocks were the elements Fire, Air, Water, and Earth. Epicurus called them "atoms", and had them indivisible, and thought their motion was the reason for large scale motion.
