It won't hurt. DON'T go into how you're actually looking for a full-time permanent position, but DO mention work situations you've been comfortable in in the past, emphasizing some particular skills.
Maybe they WILL have something suited to you more long-term; just don't put them off asking for it.

You're reading I am answering an ad for an exam proctor at a local school. It's a weekend-long gig, but pays well. Do I need to submit a cover letter if it's only a two-day long temp position?

The ad says for me to describe my work/testing experience. I am not sure if they want a formal letter or just a brief paragraph summarizing my work experience and skills.
