Think about the book and what its message is, think about what you want people to think when they pick it up, and how you are going to grab their attention etc. If you mean how can you get to be asked to design a real book cover, maybe do a graphic design course, or get involved with your local art college.

Use a picture and colors that catch your eye. Don't use more that two types of text, and try not to get a "crowded" effect. Use font sizes/styles that are easy to read but don't overwhelm your background. (Unless that is your intent.) Make sure your design is balanced, not "busy" on one side and "blank" on the other.
