I'm wearing a long, flowing, floral button-front skirt with a long flowing white slip, both ankle length. Should I avoid having buttons on the skirt undone from the hemline up?

I would say it is a fashion "don't" to show a slip of any kind, especially if it is ankle length.

It depends on the colours in the skirt. If the background to the pattern is coloured (maybe blue or pink) and the flowers white or another pale colour it would be an unusual but effective look as it would make use of the contrast. If the background of the pattern is white it will probably just look as if the buttons came undone by mistake. If its a very decorative embroidered slip you might get away with it though. Personally I think a coloured slip would be better.

It would be appropriate to open some buttons on this skirt if you were wearing a decorative petticoat underneath, but not when wearing a regular slip.

I'm of the view that long skirts and dresses can be decorated with a slip or petticoat, assuming the slip or petti itself is decorative. I have several long satin and lace petticoats, in a variety of colors, to wear under my longer skirts and dresses. The lace shows a bit under the skirt/dress hem when both are still and shows a lot more when both are in motion (I'm walking). I don't care for or wear plain slips of any color (I prefer lots of pretty lace), but if I did have one, I would not show it with my dress or skirt because it would look like I'm a slob who doesn't care that my slip is showing. On occasion I have worn shorter dresses/skirts with petticoats that push out the dress or skirt and the petti will show when I walk, sit down, etc. That can look really cute, but I've made the mistake of wearing a petticoat under a very short skirt only to realize that very little movement on my part would result in my panties showing.

If the skirt is of equal fullness to the slip there is no point in having it undone. Perhaps too, if it were undone from the hem to the waist, you would surely experience some difficulty in gathering it all up for a call of nature

I would avoid exposing the slip (actually, I would skip the slip altogether, but that's a separate discussion). Exposing the slip by unbuttoning the skirt at the hemline will not look right.

You should forgo the slip and have a few buttons open.
How many depends on your level of comfort!
