That was never really clarified, but I am pretty sure that Misty adores Ash. Whether Ash likes Misty the same way, I don't know for sure.

In an episode when a girl has a crush on Brock, Misty secretly has a crush on Ash. She says to Ash: "Maybe one day we'll be married too" Secondly,when Misty tells Brock to like the girl as well, she says something like "Trust me on this, Its better to like someone that already likes you" and this conversation goes back to "Maybe one day we'll be married too" Sorry, cant explain it properly.
