I've never played the DS version, but I can only assume it's nearly identical to the N64 version, in which different parts of the castle are accessed by unlocking doors with keys received upon defeating Bowser.

you first need at least 30 stars to enter the second floor, you need also to have unlocked mario and luigi and must have stomped on the two pillars that raise to water level of the castle(enter the door to the left of the star door in the basement to do this). After you've done all that enter the star door in the basement of the castle( you first need to have gotten the first star of DIRE, DIRE DOCKS) and enter the hole in between the DIRE, DIRE and the star door in the middle of the hallway. In there you must defeat bowser and obtain the key to enter the second floor.
