In a wet preparation, you would be able to see clumps or groups under 10X lens with 5X or 10X eyepieces (=50x or 100x). A 40X lens will enable you to distinguish cocci and bacilli, as well as basic Gram pos or neg differentiation (= 200x or 400x)but a 100x lens will be required for smaller bacteria like Haemophilus or Campylobacter and is betterlens to focus on specific morphology etc (= 500x or 1000x)
