Obviously they are looking for a yes. Generally this question is really meaning 'Why do you want this job and why do I want to employ you' Answer truthfully, or not so if you need the job, but expand on yes as to why you want it - and dont say you need money :p
-
First a positive attitude and that they can and will be on time /past jobs and why they left - experience they have and how willing they are to learn

I agree with Starson, and also -- When you respond, emphasize the professional-sounding reasons why you want it, and emphasize specific benefits you believe you can contribute IN the job. Learn everything you can about the company and the job in advance, so that you can view it as much as you can from THEIR point of view. What you know/have learned/can do adds value - talk about value to their company. Something applicants sometimes forget is that they need to differentiate themselves favorably from all the other applicants whose credentials on paper probably look just about the same as theirs.

When I ask that question, I'm looking for someone to tell me obviously "Yes" and why they are the best/only possible choice for the job. If a candidate is so submissive that they won't step forward and tell me that they SHOULD have the job, I am not terribly interested in that candidate...

im appying for this job because i want to try something that i have never done before something more challenering

They want to see if you are really interested in the content of your working day and not just after the job because of the money.
