At least 24 hours to allow the chlorine to evaporate from the water. If you are on a well, you'd better have a county extension rep test your water first unless you have your own kit. Check for the exp. date of you have one.

Until the cycling period is complete and the water tests negative for ammonia. (Ammonia will kill fish) Depends on what you used to cycle the tank and other variables, for example, a tank cycles faster in warm water than cold. The whole process can take anywhere from a few days to two or three weeks.
