The screw in the rotor doesn't hold the e-brake, it is just there for shipping the rear end assembly. You don't need it. The resistance is probably the shoe for the park brake that is in the "HAT" of the rotor. You need to see if you can find and loosen the adjuster for the park brake. It can help to turn the rotor while you are pulling it off.
