Yes, it will affect the amount of disk space available while your external drive is attached.

there is nothing wrong with mixing brands and capacities. If your computer is very old though, it might not accommodate the newer high capacity disks, or you might have to update the BIOS on your motherboard.

No problems. Just be aware an external hard-disk might be a little limited in speed by the USB 2.0 specifications, so you should avoid running stuff that will require too much disk access from it (like Games).
