The first thing that happens is the mail-man/woman comes to get the mail, and from there it is taken to the local distribution center (post-office). From there, based on the destination zip-code, computers that the USPS uses determines where it goes next. This process is usually repeated until it reaches its destination. If you are, for example, sending a letter to Washington D.C., once it reaches Washington D.C. that local post office determines which mail-man/woman runs the route that the letter's destination is on and gives it to them for them to deliver. This is just an educated guess, but I don't think I'm too far off.

Your letter is first picked up by a letter carrier who puts it in a bag and takes it to the local post office. There, all the letters are unpacked from the bags, are sorted by destination and repacked into the back of transport trucks by city of destination. These trucks make the trip from where you are to the post office closest to your intended receiver. There, the mail is unloaded and sorted again and placed into a delivery sequence where the letter is taken to the end destination and delivered.
