In replica plating, a culture with both mutants and wild type bacterium are plated on media that will allow everything to grow. This plate is called the “master plate” because the colonies on this plate will be imprinted onto many types of differential and/or selective media. After the bacteria have been allowed to grow the differences among the mutants and wild type bacteria can be observed thus allowing us to identify & separate the two. Hope this would be of help!,God bless
