Depends on the manufacturer. Some can simply be pulled off (usually you have to pull hard). Others have a small screw near the base of the handle that needs to be loosened. Lastly, some are permanent. Good luck!

Oh wait...do you mean a vehicle? If you do, you should mention which vehicle. If it's a Chevy S-10, then you need to get a crank remover. It looks like a U-shaped magnet on the end of a screwdriver. You slide the tool between the the crank and the door panel to trip the lock pin, then the crank will pull off.
