Yes.. A grounded ignition wire will cause that cylinder to misfire.

I had a car which would only die in a hard left hand corner. I traced the problem to one of the four spark plug wires which was in a well going to the plug. Since there was a little oil residue in the well only during the right condition would the plug short, and the engine die. Hey it was only a four cylinder to start with, running on three is not an option.
