The PlayStation 3's button layout is identical to the PS2 and PS one before it, with four face buttons (Triangle, Circle, Square and X), four shoulder buttons (L1, L2, R1 and R2), and the d-pad, all of which are analog (meaning if the game is programmed for it, the harder you press, the more of a response you'll get).
In most games, the main functions will almost always rest on the face buttons; Japanese games have traditionally used O as a confirmation (or accept) button and X backing out or cancelling. American games usually swap this for some crazy reason mandated by Sony Computer Entertainment America eons ago. Shoulders are most commonly used to tab between seconds of menus or as modifiers; first person shooters usually use them for primary/secondary fire and things like jumping or ducking.
It's really going to come down to the games, though, as there's no one preset for functions.
