It could be serious, but the two things that immediately come to mind are minor.

My first instinct when I hear a humming from my tower is that my fan motor is out of whack again. It's a personality quirk of my system; the HD cooler I put in uses cheap fans with non-ball-bearing motors.Check the fans.

Second is one of my drives shaking. Are all of the mounting screws tight?

If it is actually an electrical sound, a certified "60-cycle hum", it's probably safest and easiest to just replace the power supply. That's the only component that carries 60Hz AC.

My first thought here is always this:
When is the last time you opened the side of your case and blew out the dust and lint?
    1. unplug PC, take to un carpeted area, open side and use a can of air on it, CPU, fans, power supply(all vents on power supply back, bottom and outside on case side), cables, motherboard, HD, vent from front of case as this is where airflow starts, last tip case to side and blows rest dust out. if you feel you must reach inside. keep one arm on case at all times for a ground, to keep static ele. out of case.

    2. now plug back in and listen for you noise, power supply, CPU or another fan inside, HD etc. if noise is from any of these shut down and replace at once!

This is most probably the fan in the powersupply vibrating and making the case vibrate with it. If it goes away after a few minutes after you turn your PC on, I'll bet money that that's what the problem is.

If it's persistent, then check everything's screwed in properly and fastened tightly. Case and/or component vibration is the cause of most odd PC humming noise.

Of course, if it's an electrical-sounding humming noise, then I'd start to get a little worried... It could possibly be a fan with a worn down bearing or axle in a fan, but it's more likely to be something like a transformer or voltage stepper, perhaps in the powersupply. Get a technician to check your PC out if the problem persists, cause it's always good to be on the safe side. :)

My computer started humming so loudly that I thought it was the fan or something was about to explode or my computer was going to crash. Stangely enough, as soon as I touched the button to the disc drive door it stopped the loud humming! Wierd, I know...but worth a try!

my fan sounds like someone is constantly hoovering, its always does it and done it for as long as i can remember. When the computer is swiched on there is no sound, but after a minute or so you can hear the fans or what ever is making the sound starting up and staying at a constant level of sound, Its really annoying, and i would love to know why they are on all the time. The past 2 computers i have had have done exactly the same

My computer has been humming for two years and it's never been a problem. Do, however, check that wires aren't getting in the way of any fans and that screws are tight enough (but not too tight!).
