If you are using windows then nope, windows cant just be copied from one drive to another, it needs to be installed. You would have to install windows on the new drive first and then copy over any other files you needed. Most software, ie. office would be the same, you cant copy it, you have to install it.

If you used a disk cloning program AND made sure the BIOS of your machine was set to be able to boot from other devices, yes. Otherwise, no.

Nope. Your computer looks in the active partition of the hard drive for windows startup information. If that info isn't there, then Windows won't start. If you install windows on the portable drive and set your computer to boot from that drive, then yeah.
