Oh lots of fun things. First of all the vocal folds are really just two mucosal membranes that create sounds by undulating against one another. If you've lost your voice that usually means something has happened to these delicate little folds. The most common reason is vocal abuse which inflames the folds and makes them swell up so they cannot connect properly to produce sounds. The most common forms of vocal fold abuse are: yelling, smoking, coughing, drinking too much booze, dehydration.

Less common reasons for losing your voice are things like vocal nodes (which may be caused by long term vocal abuse), or acid reflux.
