We measure the temperature using the light given off by the stars. As substances get hotter the frequency of the light emitted by them increases. This leads to a shift in the color of the light from the red end of the spectrum toward the blue end. So, by examining the spectrum of a star and determining the color of light that is most strong in that spectrum, we can tell the temperature of the star.
