What difference would you see if a week old and an overnight culture of B. subtilis is stained with malachite green? Does the overnight culture even have spores?

    You need to specify the culture conditions. Nutrient agar medium and nutrient liquid medium will have different effects. Assuming that you are using nutrient liquid medium at 37 degree C, an overnight (i.e. 18 hours) culture will not have spores (save the ungerminated ones from the inoculums) but the week old ones will normally have some spores.
