You need to specify the culture conditions. Nutrient agar medium and nutrient liquid medium will have different effects. Assuming that you are using nutrient liquid medium at 37 degree C, an overnight (i.e. 18 hours) culture will not have spores (save the ungerminated ones from the inoculums) but the week old ones will normally have some spores.
What is the difference between normal flora and transient bacteria found on the skin?

Do I need a microscope to see living bacteria? I need to see the effects of antibiotics on bacteria, but can I see it without a microscope?

You're reading What difference would you see if a week old and an overnight culture of B. subtilis is stained with malachite green? Does the overnight culture even have spores?
