The glans; and the underside of the penis just below the head are the most sensitive parts of the penis. See illustration:

The outer edge of the glans, the knob on the end, and the underside of the penis - the central and smallest of the three columns that form it.

If you can find the vein that runs laterally down the shaft, 2 finger-widths from the head, there should be a soft spot there. Massaging this spot will drive guys crazy. Also near the bottom of the head, a downward pressure on the root will intensify the feeling

The most erogenous part of a man's penis is inside the penis, at the tip of
    the corpora cavernosa.

It's called the Lowndes Crown. Its pleasure nerves control a man's orgasm.

It's described in PART 2 on this webpage: http://xrl.us/ForeskinSexualFunctions

A complete description of this most erogenous part of the penis is in the book Sex As Nature Intended It on page 133. Below is a link to read the book in PDF eBook format on your computer (requires Adobe Reader or equivalent PDF doc reader installed). Very informative, fascinating reading.

Also, see more on the Lowndes Crown on page 367 of the book.

For me, and I'm circumcised, it is along the shaft about 1-1.5 inches beneath the head. It's a lot more sensitive than the head.
