The Permian-Triassic Extinction happened around 250 Million years ago. It killed nearly all marine life and 3/4 of terrestrial life. The event responsible for this is not well known--it could have been a combinations of events like asteroid impacts and major volcanism. This was the worst event to life on earth.

For the earth itself: There was one incident when a Mars-sized planetesimal collided with Earth breaking away a good chunk of the planet and forming our moon. I would say this was the worst physical blow to earth.

But the ultimate answer to your question is yet to come. It will take place in about 5 billion years from now when the sun will expand into a red giant causing the oceans to evaporate, the surface to go molten, and life to vanish off the planet. If earth doesn't get totally engulfed by the expanded sun, it will be a very barren, cold, lifeless place that will receive very little light from the remains of our sun--a white dwarf.
