The problem with this debate is that so many people have lost sight of the original intent of the First Amendment. No where does it mandate a separation between church and state. It's intent was to keep the federal government from favoring and supporting one religion above all others the way the British government still does for the Church of England. If you really want to know the intent of the First Amendment with respect to this question, then you should look at the other actions of those that wrote it. This amendment was one of the very first acts of the first Congress. Another very early act of that Congress was to establish the offices of Chaplain for both the House of Representatives and Senate (one chaplain for each house of Congress). The purpose of this office was to provide someone to offer a prayer at the beginning of each session in each of those houses. So, if the very men who wrote the First Amendment also established these very religious positions for their houses, is it really reasonable to interpret the amendments as having the intent to create a separation between church and state? It should be obvious that it was not. It has only been in recent years that certain groups in our society have started pushing for the interpretation that contradicts the obvious intent of the writers of this amendment.

So, no a national day of prayer is no more unconstitutional than Mothers' Day. There is nothing in the declaration of a national day of pray that states that the citizens of this nation must pray just as there is no requirement that we honor our mothers on Mothers' Day. As with any holiday in this nation, you are free to observe it or not. It is up to you.
