Let me preface my answer by saying this: Yes, swapping urine is illegal and unethical, however, I am in no position to judge people. That is not my job, and is reserved for only one -- and it's not me.
That being said. They cannot really tell age, but, they can tell gender. However, they won't test for gender unless they suspect urine swapping. Typically employers outsource urine testing to a clinic of some kind, and they take the sample and either stick test strips in it, or they run it thru an automated process that does the testing inside of it.
Urine can be tested in two different ways, and both tests look for the same things. They look for metabolites of the drugs ingested. The body metabolizes the drugs into "metabolites" and thats what the urine tests for.
Drug tests are semi-cheap, about 20 bucks for a 5-panel test (THC, Cocaine, Opiates, Barbituiates, and Methamphetamine).
The gender of a sample is determined by doing a hormone test. Obvisouly if your husbands sample comes back with high levels (for a male) of estrogen, then there is something medically wrong, or he swapped his sample.
The hormone test would be an additional cost, so to save costs, they wont test for gender unless they suspect he's done something he shouldn't have. They will check the temperature of the sample, and the color, if either of those is off, then they will look closer at the sample, and do extra tests that they normally would not.
Also, unless you were locked into a small closet with many people smoking, and the air in the closet was 90% smoke, second hand smoke will not contain enough THC for you to come up positive on a test.

Reading the other answers and comments I noticed that you implied he was an infrequent user. If that is the case this information may be helpful to you. If he used maybe once a week, he could flush his system in about 4-5 days. If he used twice a week, he could flush in about 7 days. If he used daily for any sort of extended period of time, it could take up to 30 days to flush his system out.
THC and its metabolites are stored in fat cells in the body. So if he is losing weight, that will cause more to be released into his urine. And also know that the more you urinate and defecate, the faster you flush your body out -- This is the only sure way to pass a test -- OTHER THAN NOT USING. So the more you eat and drink, the faster you will get your body clean. I recommend drinking at least 2 gallons of water a day along with taking B complex vitamins (or drinking propel fitness water as it contain B complex vitamins.) That amount of water will cause the amount of metabolites in his urine to be very low -- possibly below the point of detection of the tests. The B complex vitamins will help keep his urine yellowish. This is needed because if the urine is clear, the testing center may think he is trying to flush his system, thereby tampering with his sample. They could fail him for that, or make him retest, and if it was still clear, they could fail him for that.
The easiest way to know if he is going to pass or not, find a drug store or pharmacy or natural heath store that sells home urine tests. Get the ones for the substances you're worried about (Get two or three), and test his urine before the test. If he fails that test, he'll fail the test at the clinic. (The reason I said get a couple is this, start testing a day or so before the test, that way if he fails he'll have time to start drinking more water to flush.. Keep testing until you pass, that way you know you'll pass at the clinic.)
